The only knuckle conditioning I would suggest is doing clenched fist push up on a hard surface. Eventually you can go all out on a heavy bag with no gloves without tearing your skin up.
I used to punch a makiwara post wrapped with 1 layer of clothes line. I don't advise anyone to that. Did more damage than good. Knuckle push ups is all you need.

You can start doing the pushups DragonFire mentioned on carpet, then wood or tile, finally on cinderblock.
It was the rope on the makiwara that did the skin damage mentioned. If you put a pad over it (or hit a bag of beans or shot) the makiwara's a good idea. The one I hit is just plain wood with very little give.

hitting the heavy bag with wraps on does a good job. I bought wraps for five dollars at my local dicks sporting goods store, and that helped. But under no circumstances should you start hitting the heavy bag without wraps. I made that mistake. my hands were cut up so bad, by the fifth day, if i tapped the bag and thats super light tap, my hands would start flowing with blood. hten i bought the wraps and now my hands and tough but ultra sensitive at the same time. scars hurt. but i can hit really really really hard adn my hands are fine now. oh ya and if you plan on bareknuckling the bag than plan on breaking your hands a few times or more. you'll probably be able to touch a couple knuckles and feel a grinding in your knuckles as you open and close your hand. ya, then its probably broken.

lol, amen to that... Havin ex g/f issues, want her back... My sensai knows summink's up when eva i head in with crusty knuckles lol...
but yeah, this is a good question... (when distraught, i can flip, in which case i dont get round to wraps) but yeah, its better than a wall right... But whats the quickest you can start toughin up the knuckles after a tearin? -Jvr
